柑橘叶斑驳病毒(Citrus leaf blotch virus,CLBV)和柑橘碎叶病毒(Citrus tatter leaf virus,CTLV)均可侵染柑橘的不同种。为明确CLBV及CTLV在中国柑橘上的发生状况和分子特性,采用RT-PCR技术对来源于中国7个省的342份和346份柑橘样品进行检测,检出率分别为9.6%和11.0%。对14个CLBV分离物和15个CTLV分离物的外壳蛋白(Coat protein,CP)基因部分序列分析显示,CLBV分离物间cp核苷酸和编码氨基酸序列相似性分别为82.1%~100%和88.1%~100%,CTLV分离物的cp核苷酸和编码氨基酸序列相似性分别为89.5%~100%和92.5%~100%。测定的14个CLBV分离物在系统进化树中聚为2个组;而CTLV分离物分组不明显,该病毒的系统进化与寄主来源无明显关系。
Citrus leaf blotch virus (CLBV) and citrus tatter leaf virus (CTLV) can infect different species of citrus. To clarify the occurrence and molecular characteristics of CLBV and CTLV in Chinese citrus, 342 and 346 citrus samples from 7 provinces of China were detected by RT-PCR with detection rates of 9.6% and 11.0%, respectively. The partial sequence analysis of the Coat protein (CP) gene of 14 CLBV isolates and 15 CTLV isolates revealed that the similarities of cp nucleotide and encoded amino acid sequences between CLBV isolates were 82.1% -100% and 88.1 % ~ 100%. The nucleotide and amino acid sequence identities of CTLV isolates were 89.5% ~ 100% and 92.5% ~ 100%, respectively. Fourteen CLBV isolates were clustered into two groups in the phylogenetic tree. However, the CTLV isolates were not significantly grouped and the phylogenetic relationship of the virus was not significantly related to the host source.
