目的:建立红参须中人参皂甙Re的含量测定的方法,并确定最佳的色谱条件。方法:采用索氏提取法和正丁醇提取法提取人参皂甙Re。高效液相色谱法测定hanliang。色谱条件:色谱柱No-va—pak(5μm,150×3.9mm),检测波长203nm,流动相为乙腈-0.05%磷酸溶液(21:79),流速为1ml/min,保留时间为6.7min。结果:人参皂甙Re的保留时间为6.7min。线性范围:0.25μg/ml—0.5mg/min(r=0.9998);定量限20ng;回收率95%-105%。人参皂甙Re在人参须中的含量为1.72-1.84mg/g。结论:HPLC法可以很好的测定人参皂甙的含量,色谱条件具有较好的专有性、灵敏度和准确度。
Objective: To establish a method for determination of ginsenoside Re in red ginseng and determine the optimal chromatographic conditions. METHODS: Ginsenoside Re was extracted by Soxhlet extraction and n-butanol extraction. HPLC determination. Chromatographic conditions: column No-va-pak (5 μm, 150×3.9 mm), detection wavelength 203 nm, mobile phase acetonitrile-0.05% phosphoric acid solution (21:79), flow rate 1 ml/min, retention time 6.7 min. Results: The retention time of ginsenoside Re was 6.7 min. Linear range: 0.25 μg/ml-0.5 mg/min (r=0.9998); limit of quantification 20 ng; recovery 95%-105%. The content of ginsenoside Re in ginseng whisker was 1.72-1.84 mg/g. Conclusion: The HPLC method can be used to determine the content of ginsenosides. The chromatographic conditions have good specificity, sensitivity and accuracy.
