目的 探讨 4 4KD受体相关蛋白 (RAP)与脂代谢的关系。方法 制备实验性家兔高脂血症动物模型 ,运用分子生物学技术克隆RAP基因片段 ,制备GST -RAP融合蛋白 ,用单克隆技术制备抗RAP单克隆抗体rapF2 ,用Western -Blot方法及免疫组化法观察高脂血症家兔心肌、血管壁中RAP的表达及分布。结果 与对照组相比 ,高脂血症组心肌、血管壁中RAP表达增加 ,且主要分布于心脏的外膜、冠状动脉内壁与外周血管壁的内、外膜 ,并在6 7~ 94KD间出现特异表达带。结论 血液中过量的脂质及脂质代谢的紊乱可引起血管壁的病理改变并伴随RAP表达增加 ,RAP表达可能与脂质的代谢过程密切相关
Objective To investigate the relationship between 4 4KD receptor related protein (RAP) and lipid metabolism. Methods Rabbit model of hyperlipidemia in rabbits was established. RAP gene fragment was cloned by molecular biology technology to prepare GST-RAP fusion protein. Monoclonal antibody was used to prepare rapF2 anti-RAP monoclonal antibody. Western blotting and immunohistochemistry Method to observe the expression and distribution of RAP in myocardial and vascular wall of rabbits with hyperlipidemia. Results Compared with the control group, the expression of RAP in myocardial and vascular wall of hyperlipidemia group was increased, and mainly distributed in the adventitia of the heart, the inside and outside of the coronary artery wall and the peripheral blood vessel wall, and between 67 and 94KD Occurrence of specific expression zone. Conclusion Excessive lipid metabolism and lipid metabolism in the blood can cause pathological changes in the blood vessel wall accompanied by an increase in RAP expression. RAP expression may be closely related to lipid metabolism
