目的观察小鼠接种重组乙型肝炎疫苗(rHB)后,脾T淋巴细胞释放细胞因子的免疫反应。方法80只BALB/c小鼠随机分为4组,1组小鼠接种5μg佐剂,2、3、4组小鼠分别腹腔接种0·65、1·25和5·0μg的rHB,其中一半小鼠2周后加强免疫1次。分别在初次免疫后4周和加强免疫后2周,分离小鼠脾T淋巴细胞,体外用rHB刺激,检测释放IL-2及IFN-γ的水平。结果单次免疫后和加强免疫后各组IL-2和IFN-γ均显著升高,且有随剂量增加而升高的趋势。结论小鼠接种rHB后,脾T淋巴细胞分泌IL-2及IFN-γ显著增加,并与接种剂量密切相关。
Objective To observe the immune responses of splenic T lymphocytes to release cytokines after mice were vaccinated with recombinant hepatitis B vaccine (rHB). Methods Eighty BALB / c mice were randomly divided into four groups. One group of mice was inoculated with 5μg of adjuvant. The second, third and fourth groups of mice were intraperitoneally inoculated with rHB 0.65, 1.25 and 5.0 μg, respectively, of which half Mice were boosted 2 weeks later. Spleen T lymphocytes were isolated from mice at 4 weeks after primary immunization and 2 weeks after booster immunization. The levels of IL-2 and IFN-γ were measured by rHB stimulation in vitro. Results The levels of IL-2 and IFN-γ in each group were significantly increased after single immunization and after booster immunization, and tended to increase with dose. Conclusions After rHB inoculation, the levels of IL-2 and IFN-γ secreted by splenic T lymphocytes were significantly increased, which were closely related to the dose of rHB.
