目的 研究周围神经损伤后白细胞介素 - 1 β( interleukin- 1 β,IL- 1 β)对周围神经损伤后脊神经节神经元的保护作用。方法 Wistar大鼠 2 0只 ,随机分为 2组 ,每只动物均切断右侧的坐骨神经 ,以硅胶管套接于坐骨神经损伤近端 ,囊内分别给予 1 ng/ml的 IL- 1 β和 PBS液各 1 5μl,术后 2周 ,应用酶组织化学方法进行相应节段脊神经节感觉神经元 Ach E染色 ,通过标准网格法进行神经元计数并应用计算机图像分析测定胞体的直径和面积。结果 与正常侧对比 ,损伤侧脊神经节对照组的神经元胞体的直径和面积明显增加 ;实验组的神经元胞体的直径和面积增加不明显 ( P>0 .0 5) ;两组损伤侧之间的差异具有极显著性意义 ( P<0 .0 1 )。结论 周围神经损伤后 ,外源性 IL- 1 β对脊神经节内神经元胞体具有保护作用。
Objective To study the protective effect of interleukin-1β (IL-1β) on spinal ganglion neurons after peripheral nerve injury. Methods Twenty male Wistar rats were randomly divided into 2 groups. Each rat was anesthetized with 1 ng / ml IL-1 β and PBS, respectively, by sutured to the proximal sciatic nerve injury with silicone tube. Liquid, each 1 5μl, 2 weeks after surgery, the corresponding segmental ganglion neurons were stained with Ach E by enzymatic histochemical method. The neurons were counted by standard grid method and the diameter and area of the somatic body were determined by computer image analysis. Results Compared with the normal side, the diameter and area of the neuronal soma in the control group were significantly increased. The diameter and area of neuronal soma of the experimental group did not increase significantly (P> 0.05) The difference between the two groups was significant (P <0.01). Conclusion Exogenous IL-1β has a protective effect on the neuronal soma of the dorsal root ganglion after peripheral nerve injury.
