目的:建立1,8-二羟基-3-羧基蒽醌(大黄酸)的反相HPLC定量测定方法。方法:色谱条件:Kromasil C18(250mm×4.6mm,5μm),紫外检测器,检测波长431nm;甲醇-0.1%磷酸(85∶15)为流动相。结果:大黄酸在0.108~2.16μg范围内线性关系良好;回收率103.2%,RSD 1.64%。结论:方法准确,精密,重现性好,可准确测定蒂达等天然化合物试样中极性组分大黄酸的含量。
Objective: To establish a reverse-phase HPLC quantitative method for the determination of 1,8-dihydroxy-3-carboxyanthraquinone (rhein). Methods: Chromatographic conditions: Kromasil C18 (250mm × 4.6mm, 5μm), UV detector with detection wavelength of 431nm and methanol-0.1% phosphoric acid (85:15) as mobile phase. RESULTS: Rhein had a good linearity in the range of 0.108-2.16 μg with a recovery of 103.2% and a RSD of 1.64%. Conclusion: The method is accurate, accurate and reproducible. It can accurately determine the content of rhein in the natural compound samples such as TEDA.
