建立了HPLC法测定兔血浆中的劳拉西泮,考察鼻腔和静注给药后家兔体内的药动学特征。结果表明,劳拉西泮在1~75 ng/ml浓度范围内线性关系良好,精密度及回收率试验结果符合生物样品分析要求。家兔静注和滴鼻给予劳拉西泮后,tmax为(9.2±10.2)和(6.7±4.1)min,AUC0→∞为(3 386.2±520.5)和(1 693.2±818.8)ng.ml-1.min,滴鼻剂的绝对生物利用度可达到62.8%。
A HPLC method was established for the determination of lorazepam in rabbit plasma and the pharmacokinetic characteristics of rabbits after nasal and intravenous administration were investigated. The results showed that lorazepam in the range of 1 ~ 75 ng / ml linearity good, precision and recovery test results in line with the requirements of biological samples. The tmax was (9.2 ± 10.2) and (6.7 ± 4.1) min after intravenous and intranasal administration of lorazepam in rabbits, and the AUC0 → ∞ was (3 386.2 ± 520.5) and (1 693.2 ± 818.8) ng.ml- 1.min, the absolute bioavailability of nasal drops can reach 62.8%.
