目的研究环氧化酶2抑制剂塞来昔布对鼻咽癌细胞株HNE-1细胞增殖作用及对血管内皮生长因子(vascularendothelialgrowthfactor,VEGF)表达的影响。方法采用噻唑蓝法[3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide,MTT]观察塞来昔布对人鼻咽癌细胞增殖的影响,流式细胞仪分析细胞周期分布,免疫细胞化学检测VEGF蛋白表达。结果塞来昔布50μmol/L作用48h后使HNE-1细胞G0/G1期细胞显著增多(由62·13%上升至91·35%),而G2/M期和S期细胞显著减少(分别由21·59%降至3·56%和16·28%降至5·01%),细胞生长停滞于G1/S期,细胞VEGF蛋白表达明显减弱。结论塞来昔布对体外培养的鼻咽癌细胞具有明显抑制作用,使细胞生长停滞于G1/S期,且能抑制VEGF蛋白表达。
Objective To study the effect of cyclooxygenase 2 inhibitor celecoxib on the proliferation of nasopharyngeal carcinoma cell line HNE-1 and its effect on the expression of vascular endothelial growth factor (VEGF). Methods The effect of celecoxib on the proliferation of human nasopharyngeal carcinoma cells was observed by MTT assay. Cell cycle was analyzed by flow cytometry Distribution, immunocytochemistry detection of VEGF protein expression. Results After treated with 50μmol / L of celecoxib for 48h, the number of G0 / G1 phase cells increased significantly from 62.13% to 91.35%, while the cells in G2 / M phase and S phase decreased significantly From 21.59% to 3.56% and from 16.28% to 0.01%). The cell growth was arrested in the G1 / S phase and the expression of VEGF protein in the cells was significantly decreased. Conclusion Celecoxib inhibits nasopharyngeal carcinoma cells cultured in vitro, arrests cell growth in G1 / S phase and inhibits VEGF protein expression.
