目的探讨姜黄素预处理对小鼠急性脊髓损伤后神经功能的保护作用及其可能机制。方法小鼠随机分为4组,假手术组、模型组、对照组(甲强龙,40mg·kg-1)和实验组(姜黄素,200 mg·kg-1)。造模前30 min,分别腹腔注射相应药物,用改良的Allen’s法建立急性脊髓损伤模型后,进行行为学评价,用Western blot法测定脊髓核因子E2相关因子2(Nrf2)蛋白表达,用酶联免疫吸附法(ELISA)测定血清肿瘤坏死因子α(TNF-α)和白细胞介素6(IL-6)的表达。结果与模型组比较,实验组小鼠的后肢功能及斜板实验评分显著增加,Nrf2和TNF-α表达均明显改善,IL-6表达在8 h减少,但之后的时间点无明显差别。结论预防性给予姜黄素,可以通过上调Nrf2、下调炎症因子对小鼠起到神经功能的保护作用。
Objective To investigate the neuroprotective effect of curcumin pretreatment on acute spinal cord injury in mice and its possible mechanism. Methods Mice were randomly divided into 4 groups: sham operation group, model group, control group (40 mg · kg-1) and curcumin (200 mg · kg-1). The rats were injected intraperitoneally with the corresponding drugs 30 minutes before model establishment. The model of acute spinal cord injury was established by modified Allen’s method. The behavioral evaluation was made. The protein expression of nuclear factor-2 (Nrf2) in spinal cord was detected by Western blot. Serum levels of tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) were measured by immunoadsorbent assay (ELISA). Results Compared with the model group, the hindlimb function and the score of experimental mice in experimental group were significantly increased, the expressions of Nrf2 and TNF-α were significantly improved, and the expression of IL-6 was decreased at 8 h. However, there was no significant difference at later time point. Conclusion Preventive administration of curcumin can protect neurons of mice by up-regulating Nrf2 and down-regulating inflammatory cytokines.
