目的探讨信号转导和转录激活因子4(STAT4)基因多态性与类风湿关节炎发病的相关性。方法以聚合酶链反应连接酶检测反应(PCR-LDR)法对139例RA患者和139名正常对照组进行STAT4基因rs7574865位点的SNP分型,比较各基因型及等位基因频率在正常人群和RA患者中的差异。结果 RA组STAT4 rs7574865各基因型分布频率与正常对照组有明显差异(P<0.05),T型和G型等位基因频率在RA组与正常对照组比较有显著差异(P<0.01,OR1.658,95%可信区间为1.154~2.383)。结论 STAT4 rs7574865位点GT/TT基因型与RA发病具有相关性,STAT4基因rs7574865位点T等位基因为RA患者的易感基因。
Objective To investigate the relationship between the polymorphism of signal transducer and activator of transcription 4 (STAT4) gene and the pathogenesis of rheumatoid arthritis. Methods PCR-LDR was used to detect SNP genotypes of rs7574865 of STAT4 gene in 139 patients with RA and 139 healthy controls. The frequencies of genotypes and alleles were compared between normal subjects And RA patients. Results The distribution frequencies of STAT4 rs7574865 in RA group were significantly different from those in normal control group (P <0.05). The frequencies of T and G allele in RA group were significantly different from those in normal control group (P <0.01, OR1). 658, 95% confidence interval 1.154 ~ 2.383). Conclusion The GT / TT genotype of STAT4 rs7574865 is associated with the pathogenesis of RA. The T allele of rs7574865 of STAT4 gene is a susceptibility gene in patients with RA.
