目的 :研究槐果碱对感染 CVB3小鼠干扰素和肿瘤坏死因子活性的影响。方法 :制备 CVB3病毒性心肌炎小鼠模型 ,用 MTT比色分析法检测小鼠脾细胞 IFN-α与 TNF的诱生量以及血清中 型干扰素 (IFN- )和TNF水平。结果 :槐果碱 5 0 mg· (kg· d) - 1 皮下注射可提高正常小鼠血清 TNF含量。与正常对照组比较 ,病毒对照组 TNF活性明显升高 ,IFN无明显变化。槐果碱 5 0 m g· (kg· d) - 1 和 2 5 mg· (kg· d) - 1 两个剂量灌胃均明显升高 CVB3感染小鼠血清和脾细胞诱生的 IFN-α活性。槐果碱两种途径、四个剂量给药均明显降低感染小鼠血清中TNF活性 ,其中槐果碱 2 5 mg· (kg· d) - 1灌胃和槐果碱 30 m g· (kg· d) - 1皮下注射还明显降低脾细胞中的 TNF诱生量。结论 :槐果碱升高 CVB3感染小鼠的 IFN活性 ,降低 TNF的活性 ,可能是其体内抗 CVB3免疫机制之一
Objective: To study the effect of sophocarpine on the activity of interferon and tumor necrosis factor in mice infected with CVB3. METHODS: A mouse model of CVB3 viral myocarditis was prepared. MTT colorimetric assay was used to detect the amount of IFN-α and TNF induced by spleen cells in mice and serum levels of interferon (IFN-) and TNF. RESULTS: Sophocarpine 50 mg·(kg·d) - 1 was injected subcutaneously to increase serum TNF in normal mice. Compared with the normal control group, the activity of TNF in the virus control group was significantly increased, and there was no significant change in IFN. Sophocarpin 50 mg·(kg·d) - 1 and 25 mg·(kg·d) - 1 both gavage significantly increased the activity of IFN-α induced by CVB3 infected mice serum and spleen cells . Sophocarpine two routes and four doses significantly reduced the activity of TNF in the serum of mice infected with Solanol 25 mg · (kg · d) - 1 gavage and sophocarpine 30 mg · (kg · d)-1 subcutaneous injection also significantly reduced the amount of TNF induced in spleen cells. Conclusion: Sophocarpine can increase the IFN activity of CVB3 infected mice and reduce the activity of TNF, which may be one of the anti-CVB3 immune mechanisms in vivo.
