目的观察三氯化钆对半乳糖胺/脂多糖(Galn/LPS)诱导急性肝损伤发生的影响。方法雄性昆明种小鼠连续2d尾静脉分别注射GdCl3(10mg/kg)或等量的生理盐水,第3天腹腔注射Galn13mg/只及LPS3μg/只或等量生理盐水,分别于Galn/LPS或等量生理盐水注射后0.5、1、1.5、6h内眦静脉取血,测定血浆丙氨酸转氨酶(ALT)活性、肝脏组织丙二醛(MDA)、谷胱甘肽(GSH)、肿瘤坏死因子-α(TNF-α)的含量。结果经Gd-Cl3预处理动物血浆ALT活性和肝脏MDA、TNF-α含量明显下降,而GSH含量则升高。结论GdCl3可通过封闭库普弗细胞(KCs)减轻Galn/LPS诱导肝内TNF-α、活性氧(ROS)等活性物质的释放,增强肝脏抗氧化应激能力,从而保护肝脏。 Objective To observe the effect of gadolinium trichloride on the incidence of acute liver injury induced by galactosamine / lipopolysaccharide (Galn / LPS). Methods Male Kunming mice were injected with GdCl3 (10mg / kg) or normal saline on the tail vein for 2 consecutive days, and Galn13mg / L and 3μg / L or normal saline on the third day respectively. 0.5, 1, 1.5 and 6 h after the injection of saline, blood samples were collected for determination of plasma ALT, MDA, GSH and TNF - α (TNF-α) content. Results The activity of ALT and the contents of MDA and TNF-α in the liver of Gd-Cl3-treated animals decreased significantly, while the content of GSH increased. Conclusion GdCl3 can protect the liver by blocking Kupffer cells (KCs) from Galn / LPS-induced hepatic TNF-α, reactive oxygen species (ROS) and other active substances release, enhance liver anti-oxidative stress ability.
