本文研究了654-2对Ach及NE引起的兔虹膜释放PGE_2及6-keto-PGF_(1α)作用的影响。Ach及NE使虹膜释放PGs增加,654-2对Ach释放PGs的作用呈剂量依赖性抑制,当654-2浓度为3×10~(-5)mol/L时,Ach(5x10~(-5)mol/L)引起的PGE_2及6-keto-PGF_(1α)的释放量分别降低31%及30%。654-2浓度高于6x10~(-5)mol/L时显著抑制NE(5x10~(-5)mol/L)释放虹膜PGs的作用,当654-2浓度为3x10~(-4)mol/L时,使NE增加虹膜释放PGE:及6-keto-PGF_(1α)的量分别从62%及34%降至7.5%及4%(p<0.01)。654-2抑制PGs释放对其抗感染性休克等作用可能是有利的。
This study investigated the effect of 654-2 on the release of PGE_2 and 6-keto-PGF_ (1α) in rabbit iris induced by Ach and NE. Ach and NE increased the release of PGs in the iris, and the effect of 654-2 on the release of PGs from Ach was dose-dependently inhibited. When the concentration of 654-2 was 3 × 10 -5 mol / L, Ach (5 × 10 -5 ) mol / L) decreased the release of PGE_2 and 6-keto-PGF_ (1α) by 31% and 30%, respectively. The effect of NE (5x10 ~ (-5) mol / L) on the release of PGs was significantly inhibited when the concentration of 654-2 was higher than 6x10 ~ (-5) mol / L. When the concentration of 654-2 was 3x10 ~ (-4) mol / L increased NE to increase iris release PGE and 6-keto-PGF 1α levels from 62% and 34% to 7.5% and 4%, respectively (p <0.01). Inhibition of PGs release by 654-2 may be beneficial for its action against septic shock and the like.
