筛选苹果转录因子MdHB1的互作蛋白,可以为研究苹果生长发育过程中各种信号分子网络提供线索和依据。本文以苹果品种‘皇家嘎啦’为试材,应用SMART技术构建酵母双杂交cDNA文库。分析MdHB1的转录活性区域并验证其对Y2HGold的细胞毒性作用,利用诱饵载体pGBKT7-MdHB1-N进行文库筛选,获得了24个阳性克隆,测序分析后得到9个无重复的MdHB1互作蛋白,其涉及光合作用、病虫害防御、逆境胁迫等过程。
Screening of the interacting proteins of apple transcription factor MdHB1 can provide clues and basis for studying various signal molecular networks in the process of apple growth and development. In this paper, the apple variety ’Royal Gala’ was used as material to construct yeast two-hybrid cDNA library by SMART technology. The transcriptional activity region of MdHB1 was analyzed and its cytotoxicity against Y2HGold was verified. The library was screened by bait vector pGBKT7-MdHB1-N and 24 positive clones were obtained. After sequencing, 9 non-repetitive MdHB1 interacting proteins Involving photosynthesis, pest and disease defense, stress and stress and other processes.
