目的检测肝细胞癌(HCC)标本中肝细胞核因子-1α(HNF-1α)和肝细胞核因子-4α(HNF-4α)的表达,探讨其在肝癌发生、发展中的作用。方法采用反转录-聚合酶链反应(RT-PCR)和免疫组织化学方法检测HNF-1α、HNF-4αmRNA和蛋白质在肝癌组织和癌旁肝组织中的表达。结果HNF-1αmRNA在癌旁肝组织中的相对表达量0.818±0.371明显高于肝细胞癌组织0.383±0.102,差异有显著性(P<0.05)。HNF-4αmRNA在癌旁肝组织中的相对表达量0.846±0.384明显高于肝细胞癌组织0.397±0.105,差异有显著性(P<0.05)。HNF-1α和HNF-4αmRNA的表达与肿瘤分化程度有关(P<0.05)。HNF-1α蛋白在癌旁肝组织中的阳性率(92.3%)明显高于肝细胞癌组织(42.3%),差异有显著性(P<0.05)。HNF-4α蛋白在癌旁肝组织中的阳性率(96.2%)明显高于肝细胞癌组织(50.0%),差异有显著性(P<0.05)。HNF-1α和HNF-4α蛋白的表达与肿瘤分化程度有关(P<0.05)。在肝细胞癌组织中HNF-1α与HNF-4αmRNA表达水平呈负相关关系。结论HNF-1α和HNF-4α在肝细胞癌中表达下调,这可能与肝癌的发生、发展相关。
Objective To detect the expression of hepatocyte nuclear factor-1α (HNF-1α) and hepatocyte nuclear factor-4α (HNF-4α) in hepatocellular carcinoma (HCC) specimens and to explore its role in the development and progression of hepatocellular carcinoma. Methods The expression of HNF-1α, HNF-4αmRNA and protein in hepatocellular carcinoma and adjacent normal liver tissue were detected by RT-PCR and immunohistochemistry. Results The relative expression of HNF-1αmRNA in the adjacent non-cancerous liver tissue was 0.818 ± 0.371, which was significantly higher than that in hepatocellular carcinoma (0.383 ± 0.102) (P <0.05). The relative expression of HNF-4αmRNA in the adjacent liver tissue was 0.846 ± 0.384, which was significantly higher than that in hepatocellular carcinoma (0.397 ± 0.105) (P <0.05). The expression of HNF-1α and HNF-4αmRNA correlated with the degree of tumor differentiation (P <0.05). The positive rate of HNF-1α protein in paracancer liver tissue (92.3%) was significantly higher than that in hepatocellular carcinoma (42.3%), the difference was significant (P <0.05). The positive rate of HNF-4α protein in paracancer liver tissue (96.2%) was significantly higher than that in hepatocellular carcinoma (50.0%), the difference was significant (P <0.05). The expression of HNF-1α and HNF-4α protein correlated with the degree of tumor differentiation (P <0.05). HNF-1α and HNF-4α mRNA expression levels in hepatocellular carcinoma were negatively correlated. Conclusion HNF-1α and HNF-4α are down-regulated in hepatocellular carcinoma, which may be related to the occurrence and development of hepatocellular carcinoma.
