目的探讨肝脾不调型腹泻型肠易激综合征(IBS-D)动物模型的制备方法。方法为确定造模周期,32只乳鼠分为模型3天组、6天组、9天组和空白对照组;为确定实验动物体重,48只大鼠按体重分层,随机分为高、中、低体重模型组和相应空白对照组;为确定致泻方法,32只大鼠分为高乳糖组、应激组、高乳糖加应激组和空白对照组,分别比较相关各组大鼠体重、腹泻情况。确定造模方法后,16只大鼠分为模型组和空白对照组,检测5-羟色胺(5-HT)、乳酸脱氢酶(LDH)及水通道蛋白(AQP4)验证模型。结果依据造模周期实验、确定动物体重实验及不同致泻方法实验的结果,确定最佳造模方法为:实验周期6天,选取高体重大鼠为实验对象,采用高乳糖饲料加束缚应激联合致泻。模型验证实验:与空白对照组比较,模型组5-HT及LDH下降(P<0.05或P<0.01),AQP4升高(P<0.05)。结论该模型模拟临床IBS以胃肠症状为主要表现、多与情绪相关的特点,具备IBS-D病理改变,可用于相关药物的实验研究。
Objective To investigate the preparation method of IBS-D animal model of diarrhea-free type of diarrhea. Methods To determine the modeling cycle, 32 neonatal rats were divided into 3 days, 6 days, 9 days and blank control groups. To determine the body weight of experimental animals, 48 rats were divided into three groups according to body weight: Middle and low body weight model group and corresponding blank control group. To determine the diarrhea method, 32 rats were divided into high lactose group, stress group, high lactose plus stress group and blank control group. The best method of modeling was determined as follows: the experimental period was 6 days, the high-body-weight rats were selected as the experimental subjects, the high lactose diet plus restraint stress Joint diarrhea. Model validation experiments: Compared with the blank control group, 5-HT and LDH in the model group decreased (P <0.05 or P <0.01) and AQP4 increased (P <0.05). Conclusion This model simulates IBS as the main manifestation of gastrointestinal symptoms, with more emotion-related features, with IBS-D pathological changes, which can be used for experimental studies of related drugs.
