目的研究上皮-间质细胞转化相关上皮细胞标志因子在C57小鼠Lewis肺癌中的表达以及肺岩宁方对其mRNA表达的影响。方法采用实时PCR观察C57小鼠Lewis肺癌右腋下移植瘤和远处转移灶发生的肺组织中上皮型钙黏附蛋白(E-cad-herin)、α-连环蛋白(α-catenin)、β-连环蛋白(β-catenin)mRNA表达及肺岩宁方对其表达的影响。结果肺岩宁治疗组较模型组肺转移率明显降低(P<0.01);对于各组移植瘤组织中,肺岩宁治疗组及化疗阳性对照组的移植瘤组织中E-cadherin、α-catenin及β-catenin均较模型组升高(P<0.01);模型组肺组织中E-cadherin、α-catenin、β-catenin表达明显低于正常肺组织中的表达(P<0.05),而经过肺岩宁方治疗后,三个标志因子均有显著升高(P<0.05或P<0.01)。结论肺岩宁方具有上调上皮细胞标志因子的作用,其可能通过抑制上皮-间质细胞转化过程的发生而达到抗肿瘤转移的作用。
Objective To investigate the expression of epithelial-mesenchymal transition-associated epithelial cell marker in Lewis lung cancer in C57 mice and the effect of Feiyanning Recipe on its mRNA expression. Methods Real-time polymerase chain reaction (PCR) was used to observe E-cadherin, α-catenin, and β-catenin in lung tissues of C57 mice with Lewis lung cancer in right infraorbital tumor and distant metastases. Results Compared with model group, the lung metastasis rate in Feiyanning group was significantly lower than that in model group (P<0.01). For all groups of xenograft tumors, E-cadherin and α-catenin were transplanted in lung cancer group and chemotherapy positive control group. Both β-catenin and β-catenin were higher than those in the model group (P<0.01). The expression of E-cadherin, α-catenin and β-catenin in the lung tissue of the model group was significantly lower than that in the normal lung tissue (P<0.05). After treatment with Feiyan Ningfang, all three markers were significantly increased (P<0.05 or P<0.01). Conclusion FeiYan-NingFang has the effect of up-regulating epithelial cell markers, which may achieve anti-tumor metastasis by inhibiting the epithelial-mesenchymal cell transformation process.
