目的:从不同动物软骨中提纯硫酸软骨素(CS)并比较其降血压活性。方法:牛、鸡软骨经稀碱-酶解法提取,Sevag法除蛋白、季铵盐络合法分离纯化。以SPF级小鼠建立高血压动物模型进行降血压活性测试。结果:提纯的硫酸软骨素经电泳、色谱检查不含肽类、氨基酸及其他酸性粘多糖,经红外光谱检查与Sigma公司硫酸软骨素A一致。低剂量研究中牛肩胛骨硫酸软骨素(BCCS)及鸡肋骨硫酸软骨素(CCCS)均能显示出降血压的作用,并且BCCS起效快,但持续时间短;CCCS起效慢但持续时间长。结论:提取的BCCS、CCCS纯度均较高,动物实验结果显示CCCS与BCCS均具有一定的降压效果,其活性CCCS优于BCCS。
OBJECTIVE: To purify chondroitin sulfate (CS) from different animal cartilages and compare its hypotensive activity. Methods: The bovine and chicken cartilage were extracted by dilute alkali-enzymolysis method, the protein was removed by Sevag method, and the quaternary ammonium salt complexation method was used for isolation and purification. Hypertensive animal model was established with SPF mice to test hypotensive activity. RESULTS: Purified chondroitin sulfate was electrophoresed and free of peptides, amino acids and other acidic mucopolysaccharides by spectroscopic examination. The results were consistent with those of Sigma Sigma Chondroitin A by infrared spectroscopy. BCCS and chick chondroitin sulfate (CCCS) showed low blood pressure in low-dose study, and BCCS showed rapid onset but short duration, while CCCS started slowly but lasts longer. CONCLUSION: The purity of BCCS and CCCS extracted are high. The animal experiments show that both CCCS and BCCS have a certain antihypertensive effect, and the activity of CCCS is better than that of BCCS.
