将Immunocaspase-3基因转染Jurkat T淋巴细胞, 使其稳定地分泌性表达靶向促凋亡蛋白Immunocaspase-3, 间接免疫荧光检测表明该蛋白可内化进入ErbB2阳性的SKBr3乳腺癌细胞, 细胞计数结果表明乳腺癌细胞的生长和增殖受到明显抑制. 进一步将Immunocaspase-3基因克隆入逆转录病毒载体pLNCX, 转染PA317细胞进行包装, 挑选高滴度的包装细胞株收获病毒. 用收获的病毒感染刺激分裂的人外周血T淋巴细胞, 经筛选后通过尾静脉注射给荷有SKBr3乳腺癌的裸鼠, 观察到肿瘤生长明显受到抑制(抑制率 73.25%), 裸鼠存活时间较对照组延长80.95%. 免疫组织化学染色结果表明裸鼠肿瘤组织中有Immunocaspase-3蛋白分布, TUNEL检测证实肿瘤细胞发生凋亡. 上述结果表明, T淋巴细胞分泌的Immunocaspase-3在裸鼠体内可以识别和进入ErbB2阳性的乳腺癌细胞, 抑制肿瘤生长.
Immunocaspase-3 gene was transfected into Jurkat T lymphocytes to make it stably secretively express Immunocaspase-3 which is a pro-apoptotic protein. Indirect immunofluorescence assay showed that the protein could be internalized into ErbB2-positive SKBr3 breast cancer cells.
