应用大鼠离体工作心脏模型研究黄芪皂甙对心肌缺血-再灌注损伤的影响,观察再灌注后心脏血流动力学的变化及其与氧自由基的关系,探讨黄芪皂甙的作用及其作用机制。结果:缺血-再灌注组(I/R组)AP、LVSP、+dp/dtmax、-dp/dtmax显著下降(P<0.05),而LVEDP、T值显著增高(P<0.05),CO、SV、CI、SI显著减小(P<0.05,或P<0.01),用药组(AMS组)收缩舒张功能均得到良好的恢复。CF显著下降(P<0.05),用药组则有显著扩冠作用(P<0.05)。心肌组织SOD含量明显下降,LPO含量增高,OFR波谱信号增强,用药组SOD含量增高,LPO含量、OFR波谱信号降低,与缺血-再灌注组比较均P<0.05。提示:黄芪皂甙抗氧自由基的作用可能是其改善心脏血流动力学,抗心肌缺血-再灌注损伤的重要机制。
To study the effect of astragalus saponins on myocardial ischemia-reperfusion injury in rat isolated heart working model, observe the changes of cardiac hemodynamics after reperfusion and its relationship with oxygen free radicals, and discuss the effects and effects of astragalus saponins mechanism. RESULTS: In the ischemia-reperfusion group (I/R group), AP, LVSP, +dp/dtmax, and -dp/dtmax were significantly decreased (P<0.05), while LVEDP and T were significantly increased (P<0.05). SV, CI and SI were significantly decreased (P<0.05, or P<0.01). The systolic and diastolic function of the medication group (AMS group) was well restored. CF significantly decreased (P<0.05), while the medication group had significant coronary stenosis (P<0.05). Myocardial tissue SOD content was significantly decreased, LPO content increased, OFR spectrum signal was enhanced, the SOD content in the medication group was increased, LPO content, OFR spectrum signal was decreased, compared with the ischemia-reperfusion group, P<0.05. It is suggested that the anti-oxidative effects of Astragalus membranaceus may be an important mechanism to improve cardiac hemodynamics and anti-myocardial ischemia-reperfusion injury.
