目的:探讨黄芪提取物中两种指标性成分毛蕊异黄酮苷与黄芪甲苷,采用反相高效液相(RP-HPLC)色谱法的测量方法。方法:使用Nobel Kromasil C8,4.6毫米×500毫米×5微米的色谱柱,以每分钟1毫升的流动相洗脱,其流动相按照适当的比例制备,以乙腈兑纯净水制备而成,采用的检测波长分别为240纳米和200纳米。结果:黄芪提取物中的黄芪甲苷毛蕊异黄酮苷这两种成分的峰面积与其进样量含有良好的线性关系,并且它们的相关系数均超过0.9995,仪器的精密度的标准差不超过2%,平均回收率均大于98%。结论:反相高效液相色谱法测定黄芪提取物中的毛蕊异黄酮苷与黄芪甲苷两种指标性成分,速度快,操作简单,回收率高,是定量芪提取物中这两种指标性成分的有效方法。
OBJECTIVE: To investigate the two components of calycosin and astragaloside in astragalus extract, and determine the content by RP-HPLC. Method: A Nobel Kromasil C8, 4.6 mm × 500 mm × 5 μm column was used to elute with a mobile phase of 1 ml per minute. The mobile phase was prepared in the appropriate ratio and acetonitrile was prepared against purified water. Detection wavelength of 240 nm and 200 nm respectively. Results: The peak areas of the two constituents of Astragalus membranaceus in the Astragalus membranaceus extract had a good linear relationship with the injection volume, and their correlation coefficients exceeded 0.9995. The standard deviation of the precision of the instrument was no more than 2% , The average recovery rate is greater than 98%. Conclusion: RP-HPLC method for the determination of two components of calycosin and astragaloside in Astragalus extract is fast, easy to operate and high in recovery rate. It is the two index components of Astragalus extract Effective method.
