齐墩果酸通过线粒体途径诱导原髓细胞白血病细胞株细胞凋亡

目的探讨原髓细胞白血病细胞株HL-60细胞半胱氨酸蛋白酶(Caspase)-9、Smac/DIABLO、细胞色素C(Cyt-c)和线粒体凋亡诱导蛋白(Bid)表达及齐墩果酸(OA)对HL-60细胞线粒体凋亡信号通路的干预作用。方法细胞分瓶培养,实验设空白对照组和12、24、48 h的OA用药组,收集细胞,提取细胞总蛋白、线粒体蛋白和去线粒体细胞浆蛋白,Western印迹法分别检测Caspase-9、Bid、Smac/DIABLO和Cyt-c的表达。结果OA引起HL-60细胞质中Cyt-c和Smac/DIABLO含量增加,并引起Caspase-9的激活,线粒体Bid蛋白含量增加。结论 OA可以通过线粒体凋亡信号通路诱导HL-60细胞凋亡。 Objective To investigate the expression of Caspase-9, Smac / DIABLO, Cyt-c and Bid in mitochondria of HL-60 cells and the effects of oleanolic acid (OA) on the mitochondrial apoptosis signal pathway in HL-60 cells. Methods The cells were divided into bottles and cultured. The blank control group and OA treatment groups for 12, 24, and 48 h were set up. Cells were collected for total cellular protein, mitochondrial protein and mitochondrial cytosolic protein. Western blotting was used to detect Caspase-9, Bid , Smac / DIABLO and Cyt-c expression. Results OA induced the increase of Cyt-c and Smac / DIABLO levels in HL-60 cytoplasm, and caused the activation of Caspase-9 and the increase of Bid protein in mitochondria. Conclusion OA can induce apoptosis of HL-60 cells through mitochondrial apoptosis signaling pathway.
