目的探讨不同抗生素在不同时段对创伤弧菌(Vv)感染小鼠心肌的保护作用及观察小鼠心肌超微结构的动态变化。方法将100只清洁级ICR小鼠分为保护Ⅰ组(所用药物含氧氟沙星、异丙嗪、地塞米松等);保护Ⅱ组(所用药物含氧氟沙星、异丙嗪等);保护Ⅲ组(所用药物含亚胺培南、异丙嗪、地塞米松等);保护Ⅳ组(所用药物含克林霉素、异丙嗪、地塞米松等)和阳性对照Ⅴ组,每组20只。Ⅰ~Ⅴ组每只小鼠接种0.2mlVv菌液(约4.4×106菌群数ml),1h后Ⅰ~Ⅳ组肌肉注射相应药物。接着在3、6、12h取各组小鼠心肌组织进行超微结构观察并比较。结果含有该菌敏感抗生素的Ⅰ~Ⅲ组在接种Vv后8h及24h小鼠存活数均显著高于Ⅴ组和Ⅳ组(P<0.01);Ⅰ~Ⅲ组心肌的超微结构损伤较之Ⅴ组和Ⅳ组有较显著的改善,而Ⅳ组小鼠的心肌损伤与Ⅴ组接近。结论敏感抗生素氧氟沙星或亚胺培南与异丙嗪以及地塞米松等联合应用能提高Vv感染后小鼠的生存率,并改善其心肌的损害。
Objective To investigate the protective effects of different antibiotics on myocardium infected with Vibrio vulnificus (Vv) in different time periods and to observe the dynamic changes of myocardial ultrastructure in mice. Methods 100 clean-grade ICR mice were divided into protection group Ⅰ (ofloxacin, promethazine, dexamethasone and so on); group Ⅱ (drugs ofloxacin, promethazine and so on) ; Group Ⅲ was protected (the drugs used included imipenem, promethazine, dexamethasone, etc.); group Ⅳ (drug used including clindamycin, promethazine, dexamethasone, etc.) and positive control group Ⅴ, Each group of 20. Each group Ⅰ ~ Ⅴ was inoculated with 0.2 ml Vv bacteria (about 4.4 × 10 6 bacterial colonies) and each group Ⅰ ~ Ⅳ was intramuscularly injected with the corresponding drugs after 1 hour. Then at 3, 6, 12h, the myocardial tissue of the mice in each group was observed and compared by ultrastructure. Results The viability of mice in group Ⅰ ~ Ⅲ containing Vibrio vulgaris at the 8th hour and the 24th hour after vaccination were significantly higher than those in group Ⅴ and Ⅳ (P <0.01). The ultrastructural damage of myocardium in group Ⅰ ~ Ⅲ was more severe than that of Ⅴ Group Ⅳ and Group Ⅳ had a significant improvement, while the group Ⅳ myocardial injury and Ⅴ group close. Conclusions The combination of ofloxacin or imipenem, a sensitive antibiotic, with promethazine and dexamethasone can improve the survival rate of mice after Vv infection and improve their myocardial damage.
