目的:探讨钙通道阻断剂硝苯地平的血迷路屏障通透性,建立高效液相色谱仪测定豚鼠外淋巴液硝苯地平浓度的方法。方法:豚鼠腹腔注射硝苯地平1周采集外淋巴液,应用反相高效液相色谱法(reversedphasehigh performanceliquidchromatography, RT HPLC)测定外淋巴液硝苯地平的浓度。结果:本法的灵敏度为5ng/ml。在浓度为50ng/ml时的平均回收率为98%。测定的相对偏差为8. 56%。外淋巴液硝苯地平的浓度为50. 2~124. 2ng/ml。结论:硝苯地平可以通过血迷路屏障,浓度随给药时间变化。本实验方法简便、快捷、准确可靠,可有效用于豚鼠外淋巴液硝苯地平的药理学研究。
OBJECTIVE: To investigate the permeability barrier of calcium channel blocker nifedipine and to establish a HPLC method for the determination of nifedipine in guinea pig perilymph. Methods: Peritoneal nifedipine was injected intraperitoneally for 1 week in guinea pigs. Peripheral lymphadenitis was measured by reversed-phase high performance liquid chromatography (RT-HPLC). Results: The sensitivity of this method is 5ng / ml. The average recovery at a concentration of 50 ng / ml was 98%. The relative deviation measured was 8.56%. Peripheral lymphatic fluid nifedipine concentration of 50.2 ~ 124 2ng / ml. Conclusion: Nifedipine can pass through the blood-labyrinth barrier and the concentration changes with time. The experimental method is simple, fast, accurate and reliable, which can be effectively applied to the pharmacological study of nifedipine in guinea pig perilymph.
