[目的]用可视化的方法研究光活化物质α-三联噻吩(α-T)对离体培养的草地贪夜蛾卵巢细胞系Sf9细胞(Sf9)的损伤机理。[方法]用Cell Counting Kit-8法测定α-T对Sf9的细胞毒力,通过流式细胞仪、激光扫描共聚焦显微镜观察α-T在Sf9细胞内的吸收、分布和光照损伤的影响。[结果]在黑暗和光照条件下,α-T处理Sf9 24 h的IC50值分别为66.359、0.003 mg/L;α-T能够跨越细胞膜在细胞质内随机分布但不能跨越核膜进入细胞核,经光照之后,细胞大小没有明显变化,形态异常,细胞膜出现褶皱,细胞核体积明显胀大。[结论]光活化物质α-T对Sf9的增殖抑制效果显著,主要损伤细胞核和细胞膜。
[Objective] The research aimed to study the damage mechanism of light-activated substance α-tris-thiophene (α-T) on Sf9 cells (Sf9) cultured in vitro. [Method] Cytotoxicity of α-T to Sf9 was measured by Cell Counting Kit-8. The effects of α-T on the absorption, distribution and light damage of Sf9 cells were observed by flow cytometry and laser scanning confocal microscopy. [Result] The IC50 values of α-T treated Sf9 for 24 h were 66.359 and 0.003 mg / L under dark and light conditions respectively. Α-T was able to cross the cell membrane in the cytoplasm but could not cross the nuclear membrane into the nucleus. After that, there was no significant change in cell size, abnormal morphology, folds in cell membrane, and significant enlargement of nucleus volume. [Conclusion] The photoactivatable substance α-T can significantly inhibit the proliferation of Sf9 and mainly damage the nucleus and cell membrane.
