目的:探讨泻肝凉血解毒方对豚鼠银屑病样皮损IL-17和IL-23水平的影响。方法:5%普萘洛尔霜剂外涂豚鼠耳背皮肤,制备豚鼠银屑病模型。造模豚鼠随机分为3组,即模型组、阿维A胶囊组和泻肝凉血解毒方组,各10只;另取10只未造模豚鼠为空白组。阿维A胶囊组和泻肝凉血解毒方组分别以相应药液灌胃,空白组、模型组用生理盐水灌胃,连续4周。干预后光镜下观察各组豚鼠局部皮损病理变化,免疫组化法检测皮损组织IL-17和IL-23的水平,ELISA法检测豚鼠血清IL-17和IL-23水平。结果:豚鼠耳部皮肤HE染色显示,造模部位可见典型的角化过度、角化不全、棘层肥厚、真皮毛细血管扩张,有较明显的单个核细胞浸润;经泻肝凉血解毒方和阿维A胶囊治疗后,皮损组织病理显著改善。免疫组化结果显示模型组IL-17和IL-23的表达明显高于空白组,用药组IL-17和IL-23的表达均显著低于模型组;阿维A组IL-17和IL-23表达与泻肝凉血解毒方组比较差异有统计学意义(P<0.05)。ELISA检测结果显示,模型组IL-17和IL-23的水平明显高于空白组,用药组IL-17和IL-23的水平均显著低于模型组;阿维A组IL-17、IL-23水平与泻肝凉血解毒方组比较差异有统计学意义(P<0.05)。结论:泻肝凉血解毒方可以降低普萘洛尔致豚鼠银屑病样皮损IL-17和IL-23的水平。
Objective: To investigate the effect of Xiegan Liangxue Jiedu Fang on the levels of IL-17 and IL-23 in guinea pig psoriasis-like lesions. Methods: A guinea pig psoriasis model was prepared by applying 5% propranolol cream on the dorsal skin of guinea pigs. The model guinea pigs were randomly divided into three groups: model group, Avia A capsule group and Xiegan Liangxue Jiedu Decoction group, each with 10 mice; another 10 non-model guinea pigs were selected as blank group. Avia A capsule group and Xiegan cooling blood detoxification group were the corresponding liquid gavage, the blank group, the model group with normal saline gavage for 4 weeks. After the intervention, the pathological changes of the local lesions in guinea pigs were observed under light microscope. The levels of IL-17 and IL-23 in the skin lesions were detected by immunohistochemical method. The levels of IL-17 and IL-23 in guinea pig serum were measured by ELISA. Results: Hematoxylin and eosin (HE) staining of the ear skin of guinea pigs showed typical hyperkeratosis, keratosis, acanthosis, dermal capillaries dilatation and obvious infiltration of mononuclear cells. Avian A capsule treatment, the skin lesions significantly improved pathology. The results of immunohistochemistry showed that the expressions of IL-17 and IL-23 in the model group were significantly higher than those in the blank group. The expressions of IL-17 and IL-23 in the model group were significantly lower than those in the untreated group. 23 expression and Xiegan Liangxue Xiedu Fang group differences were statistically significant (P <0.05). The results of ELISA showed that the levels of IL-17 and IL-23 in the model group were significantly higher than those in the blank group. The levels of IL-17 and IL-23 in the model group were significantly lower than those in the model group. 23 levels and Xiegan Hemostatic Detoxification Fang group differences were statistically significant (P <0.05). Conclusion: Xiegan Liangxue Jiedu Fang can reduce the levels of IL-17 and IL-23 in the skin lesions induced by propranolol in guinea pigs.
