目的探讨胃组织钠氢交换蛋白1(NHE1)检测在早期胃癌诊断中的作用。方法选择2007年5月—2015年5月期间于浙江省平阳县人民医院接受手术的胃癌患者、胃镜检查中取得的胃黏膜组织共50例,人胃癌细胞株SGC-7901和人正常黏膜细胞株GES-1,比较NHE1的mRNA表达以及蛋白质表达情况。观察NHE1特异性阻断剂EIPA对SGC-7901胃癌细胞株的影响。结果胃癌样本组织及SGC-7901细胞株中NHE1的mRNA表达明显高于胃正常黏膜组织和GES-1胃黏膜细胞(均P<0.05)。胃正常黏膜组织中NHE1的IOD值与GES-1细胞株中NHE1的IOD值差异无统计学意义(P>0.05),胃癌组织中NHE1的IOD值与SGC-7901细胞株中NHE1的IOD值差异无统计学意义(P>0.05),胃癌组织中的NHE1的IOD值高于正常黏膜组织中NHE1的IOD值(P<0.05),SGH-7901细胞株的NHE1的IOD值高于GES-1细胞株中的NHE1的IOD值(P<0.05)。采用不同浓度EIPA处理SGC-7901,24 h后测定OD值,随着EIPA浓度增加,SGC-7901的OD值呈现明显下降的趋势,在EIPA浓度不超过30μm/ml的范围内具有较好的剂量-效应关系。结论胃癌患者生存率的提高有赖于早期的诊断和早期正确的治疗,胃组织NHE1蛋白检测用于辅助诊断胃癌或癌前病变有一定的临床意义。
Objective To investigate the role of NHE1 in the diagnosis of early gastric cancer. Methods From May 2007 to May 2015, gastric cancer patients undergoing gastroscopy in Pingyang County People’s Hospital of Zhejiang Province were included in this study. Gastric mucosa tissues were obtained from gastric mucosa in 50 cases, human gastric cancer cell line SGC-7901 and human normal mucosa cell line GES-1, comparing NHE1 mRNA expression and protein expression. To observe the effect of EIPA, a NHE1-specific antagonist, on SGC-7901 gastric cancer cell lines. Results The mRNA expression of NHE1 in gastric cancer tissue and SGC-7901 cell line was significantly higher than that in gastric normal mucosa and GES-1 gastric mucosa (all P <0.05). There was no significant difference in the IOD value of NHE1 between normal gastric mucosa and NHE1 in GES-1 cell line (P> 0.05). The difference between IOD value of NHE1 in gastric cancer tissue and NHE1 in SGC-7901 cell line The IOD value of NHE1 in gastric cancer tissue was higher than that in normal mucosa tissue (P <0.05), and the IOD value of NHE1 in SGH-7901 cell line was higher than that in GES-1 cells IOD values of NHE1 in the strains (P <0.05). The OD value of SGC-7901 treated with different concentrations of EIPA for 24 h was measured. The OD value of SGC-7901 decreased significantly with the increase of EIPA concentration. The optimal concentration of EIPA was 30 μm / ml - the relationship between effects. Conclusion The improvement of survival rate in patients with gastric cancer depends on early diagnosis and early correct treatment. The detection of NHE1 protein in gastric cancer may be of clinical significance in the diagnosis of gastric cancer or precancerous lesions.
