目的:探求光敏剂5-氨基酮戊酸(ALA)口服后在膀胱中的代谢,寻求光敏诊断最佳的ALA口服剂量及时间。方法:予昆明小鼠口服不同剂量的ALA(20mg/kg、40mg/kg、60mg/kg8、0mg/kg 4个组别),不同时间(1小时、2小时、4小时、6小时、8小时、10小时、12小时、24小时、48小时)以激光诱导荧光OMA检测系统测定膀胱卟啉和组织荧光强度,并经数据处理观察ALA不同时间在膀胱中的代谢状态。结果:膀胱卟啉荧光峰值除以组织荧光峰值的比值在口服ALA40 mg/kg后1小时最大。结论:从实验数据分析口服ALA作OMA荧光光谱分析以40mg/kg为最佳检测剂量,最佳检测时间为口服ALA后1小时。
OBJECTIVE: To investigate the metabolism of 5-aminolevulinic acid (ALA) in the bladder after oral administration and to seek the optimal dose and time of oral ALA for photosensitization. Methods: Kunming mice were orally given different doses of ALA (20mg / kg, 40mg / kg, 60mg / kg and 0mg / kg) for different time (1 hour, 2 hours, 4 hours, 6 hours, 8 hours , 10 hours, 12 hours, 24 hours and 48 hours). Fluorescence-induced fluorescence OMA detection system was used to determine the fluorescence intensity of porphyrin and tissue. The data of ALA metabolism in the bladder were observed. Results: The ratio of fluorescence peak of porphyrin to the fluorescence peak of tissue was the highest at 1 hour after ALA 40 mg / kg. Conclusion: Oral fluorescence spectrometry (OMA) analysis of oral ALA from experimental data is the best detection dose of 40mg / kg, the best detection time is 1 hour after oral administration of ALA.
