目的观察甲醛固定的H22肿瘤细胞联合IL-2对小鼠H22肝癌细胞腹部移植瘤生长的影响。方法20只小鼠,均采用股部皮下1×105个H22细胞的方法建立小鼠H22肝癌皮下移植瘤模型。将其随机分为A、B、C、D组,各5只。肿瘤直径超过1 cm后A、B、C、D组分别瘤内注射生理盐水、甲醛固定H22细胞、IL-2、甲醛固定H22细胞+IL-2各0.5 mL。H22细胞浓度为106/mL,IL-2浓度为8×103IU/mL。每3d注射1次,共注射4次。治疗结束处死动物,取瘤称重,并计算抑瘤率。采用免疫组化SABC法对肿瘤组织染色,光景下观察瘤内CD8+T淋巴细胞表达情况。结果 A、B、C、D组肿瘤治疗后质量分别为(6.683±1.102)g、(2.851±0.223)g、(2.282±0.443)g、(1.052±0.225)g,B、C、D组与A组相比明显降低,D组与B、C组相比明显降低(P均<0.05)。B、C、D组抑瘤率分别为51.58%、60.54%、80.96%。D组小鼠瘤体内大量肿瘤细胞变性和坏死,且瘤内有大量CD8+T淋巴细胞浸润,阳性产物主要呈现在细胞膜上并被染成棕褐色。B、C组内仅见少量CD8+T淋巴细胞。结论甲醛固定的H22细胞联合IL-2具有延迟荷瘤小鼠H22肝癌形成,限制生长的作用。
Objective To observe the effect of formaldehyde-fixed H22 tumor cells combined with IL-2 on the growth of transplanted tumors of mouse H22 hepatoma cells. Methods Twenty mice were subcutaneously implanted with 1×105 H22 cells in the femoral head to establish a subcutaneous xenograft model of H22 hepatoma in mice. They were randomly divided into groups A, B, C, and D, with 5 rats in each group. After the tumor diameter exceeded 1 cm, 0.5 mL of normal saline, formalin-fixed H22 cells, IL-2, formaldehyde-fixed H22 cells and IL-2 were intratumorally injected into groups A, B, C, and D, respectively. The H22 cell concentration was 106/mL and the IL-2 concentration was 8 x 103 IU/mL. Each 3d injection, a total of 4 injections. Animals were sacrificed at the end of treatment, tumors were weighed, and tumor inhibition rates were calculated. The tumor tissue was stained by immunohistochemical SABC method and the expression of CD8+ T lymphocytes in the tumor was observed under light conditions. Results After treatment, the mass of tumors in groups A, B, C, and D was (6.683±1.102) g, (2.851±0.223) g, (2.282±0.443) g, (1.052±0.225) g, respectively, in groups B, C, and D. Compared with group A, the group A was significantly lower than group B and C (P<0.05). The tumor inhibition rates in groups B, C, and D were 51.58%, 60.54%, and 80.96%, respectively. In group D, a large number of tumor cells were degenerated and necrotic in tumor cells, and a large number of CD8+ T lymphocytes infiltrated in the tumors. The positive products mainly appeared on the cell membrane and were stained in brown. Only a small amount of CD8+ T lymphocytes were seen in groups B and C. Conclusion Formaldehyde-fixed H22 cells combined with IL-2 have the effect of delaying the formation of H22 hepatocarcinoma in tumor-bearing mice and limiting the growth.
