目的观察高糖环境下豚鼠膀胱Cajal间质细胞(interstitial cells of Cajal,ICCs)内钙离子荧光及其超微结构的改变,探讨糖尿病膀胱(DCP)的发病机制。方法采用原代培养,激光共聚焦、透射电镜观察5、10、15mmol/L葡萄糖浓度培养24、72h的ICCs,扫描记录胞内荧光强度值,观察其超微结构。结果糖浓度增高培养时间延长,ICCs内钙离子荧光值升高,胞内线粒体大小不等、数量减少、空泡样变甚至溶解,胞质广泛溶解,突起消失。结论高糖环境对豚鼠膀胱ICCs电生理学、超微结构造成显著影响,这种改变可能是造成DCP发病的重要原因之一。
Objective To observe the change of calcium ion fluorescence and ultrastructure in interstitial cells of Cajal (ICCs) in guinea pig under high glucose condition and to explore the pathogenesis of diabetic bladder (DCP). Methods Primary culture, confocal laser scanning microscopy and transmission electron microscopy were used to observe the ultrastructures of ICCs cultured in glucose concentrations of 5, 10 and 15 mmol / L for 24 and 72 h respectively. Results When the concentration of glucose increased, the culture time was prolonged, the fluorescence of calcium ion in ICCs increased, the number of mitochondria increased, the number of mitochondria decreased, vacuoles changed or even dissolved, and the cytoplasm became widely dissolved and the processes disappeared. Conclusion The hyperglycemic environment has a significant effect on the electrophysiology and ultrastructure of ICCs in guinea pig bladder. This change may be one of the important causes of DCP.
