以延边“苹果梨”果实为试材,采用同源克隆的试验方法,研究了果实着色过程的转录基因PyMYB114,并对其进行生物信息学分析。结果表明:PyMYB114的cDNA全长为744bp,其包含1个完整的编码201个氨基酸的ORF,其编码蛋白的分子量约为23.48kDa,理论等电点(pI)为9.57。同源性分析表明,PyMYB114基因与白梨的MYB114基因核酸序列的相似性高达99%。系统进化树表明,PyMYB114编码的氨基酸和白梨的MYB114编码的氨基酸在进化上亲缘关系最近。
Taking Yanbian “apple pear ” fruit as test material, the homologous cloning method was used to study the transcriptional gene PyMYB114 in fruit coloring and its bioinformatics analysis. The results showed that the full-length cDNA of PyMYB114 was 744bp and contained a complete ORF encoding 201 amino acids with a molecular weight of about 23.48 kDa and a theoretical isoelectric point (pI) of 9.57. Homology analysis showed that the similarity of PyMYB114 gene to MYB114 gene of white pear was as high as 99%. The phylogenetic tree indicated that the amino acids encoded by PyMYB114 and the amino acids encoded by MYB114 of Baizhu were most phylogenetically related.
