目的探讨胎盘人激活转录因子3(ATF3)的表达与子痫前期的关系。方法分别应用实时荧光定量-PCR(q RT-PCR)、免疫组化和Western Blot检测35例正常妊娠和38例子痫前期胎盘上ATF3 m RNA和蛋白的表达水平。结果正常妊娠组和子痫前期组胎盘ATF3 m RNA表达水平分别为1.02±0.21和1.78±0.28,两组比较,差异有统计学意义(P<0.001);ATF3蛋白在子痫前期胎盘表达升高(P<0.001);并且重度子痫前期胎盘ATF3的表达水平(189.0±62.8)明显高于轻度子痫前期(108.0±25.1,P<0.05)。结论子痫前期胎盘组织中异常表达的ATF3可能参与了子痫前期的发病。
Objective To investigate the relationship between the expression of human placental activating transcription factor 3 (ATF3) and preeclampsia. Methods The expression of ATF3 m RNA and protein in 35 cases of normal pregnancy and 38 cases of preeclampsia placenta were detected by qRT-PCR, immunohistochemistry and Western Blot respectively. Results The levels of ATF3 m RNA in placenta of normal pregnancy group and preeclampsia group were 1.02 ± 0.21 and 1.78 ± 0.28, respectively (P <0.001). The expression of ATF3 protein in preeclampsia placenta increased P <0.001). The expression of ATF3 in severe preeclampsia (189.0 ± 62.8) was significantly higher than that in mild preeclampsia (108.0 ± 25.1, P <0.05). Conclusion Abnormal expression of ATF3 in preeclampsia placenta may be involved in the pathogenesis of preeclampsia.
