本实验成功地以决明的胚轴、子叶诱导出愈伤组织。实验表明,MS比B5更利于愈伤组织诱导。正交试验表明,添加1mg/L的IAA、6-BA、GA3可明显促进愈伤组织的生长;添加1mg/L的NAA、2,4-D可抑制其生长;添加1mg/L的6-BA、2,4-D对抑制愈伤组织的分化有明显的效应。适合愈伤组织生长及维持分化率低水平的蔗糖浓度为2%~3%。愈伤组织的诱导及生长周期为30d左右。
This experiment successfully induced the callus from the cotyledons and cotyledons of Cassia. Experiments show that MS is more conducive to callus induction than B5. Orthogonal experiments showed that the addition of 1 mg/L IAA, 6-BA and GA3 could significantly promote the growth of callus; adding 1 mg/L NAA and 2,4-D could inhibit its growth; adding 1 mg/L 6- BA and 2,4-D have obvious effects on inhibiting the differentiation of callus. Sucrose concentrations of 2% to 3% are suitable for callus growth and maintenance of low differentiation rates. Callus induction and growth cycle is about 30d.
