目的:探讨同一生物组织在组织、细胞和细胞膜不同层面的傅立叶变换红外光谱(FTIR)谱图变化规律.方法:将提取肿瘤组织和瘤旁正常组织的细胞膜及提取细胞膜后剩余成份(沉淀物),于100kg/cm2下8min制成溴化钾压片行FTIR检测,指认其特征光谱并将细胞膜、沉淀物、细胞株及组织的FTIR谱图进行对比分析.结果:细胞膜与沉淀物的FTIR谱图相比,蛋白和脂类的吸收峰发生显著变化,核酸的吸收峰在细胞膜消失而在沉淀物中增强;细胞膜的FTIR谱图与组织、细胞株相比,细胞膜的蛋白质二级结构发生了明显改变,脂类吸收峰增强;沉淀物的FTIR谱图与组织、细胞株相比,蛋白二级结构发生了明显改变,脂类吸收峰增强,但与组织谱图一致;癌组织中细胞膜、沉淀物及组织水平的相对强度I1460/I1400均小于正常组织.结论:同一组织不同层面的FTIR谱图变化不全一致;肿瘤组织中脂类含量减少,但细胞膜上的脂类含量增加,肿瘤组织及肿瘤组织细胞膜的蛋白质二级结构发生了明显改变;1460cm-1和1400cm-1可作为FTIR用于肿瘤诊断的一个可靠指标.

The cell membrane, The relative intensity I1460 / I1400 of sediment and tissue level were less than that of normal tissue.Conclusion: The changes of FTIR spectra in different layers of the same tissue were not consistent, and the content of lipid in tumor tissue was decreased , But the lipid content of the cell membrane increased, the tumor tissue and tumor cell membrane protein secondary structure has undergone significant changes; 1460cm-1 and 1400cm-1 can be used as a FTIR tumor diagnosis a reliable indicator.
