目的建立马钱子药材指纹图谱,并采用化学计量学方法和双标线性校正法进行研究。方法采用HPLC建立马钱子指纹图谱,使用相似度分析、二维聚类分析和主成分分析进行质量评价,并使用双标线性校正法进行色谱峰定性。结果 10批次马钱子药材选定14个色谱峰作为指纹图谱的共有峰,进一步选择马钱苷酸、绿原酸、士的宁和马钱子碱4个色谱峰作为指纹图谱的特征峰。经化学计量学评价,可对其产地进行初步分类。双标线性校正法预测保留时间的精度优于相对保留时间法。结论本实验建立的指纹图谱有较强的针对性,可用于马钱子的质量评价。与相对保留时间法相比,双标线性校正法预测保留时间结果的准确度更高,色谱柱的适用范围更广。
Objective To establish the fingerprints of Strychnos nux vomica and to study it by the method of chemometrics and double-standard linear calibration. Methods The HPLC fingerprint of Strychnos nux vomica was established by HPLC. The quality was evaluated by similarity analysis, two-dimensional cluster analysis and principal component analysis. The chromatographic peaks were characterized by double-standard linear calibration. Results 14 lots of chromatographic peaks of 10 batches of Strychnos nux-vomica were chosen as the common peaks of the fingerprints. Four peaks of ramiglotic acid, chlorogenic acid, strychnine and strychnine were further selected as the characteristic peaks of the fingerprints . After the chemometric evaluation, the initial classification of their origin. The accuracy of the double standard linear calibration method to predict the retention time is better than the relative retention time method. Conclusion The fingerprints established in this experiment have strong pertinence and can be used to evaluate the quality of Strychnos nux vomica. Compared with the relative retention time method, the double standard linear calibration method can predict the result of retention time with higher accuracy and wider scope of application.
