目的建立高效液相色谱含量测定方法,替代原薄层扫描法测定三金胶囊中积雪草苷的含量,以更好地控制该制剂质量。方法色谱柱为Spherisorb ODS C18柱(250 mm×4.6 mm,5μm)柱,流动相为乙腈-2 mmol/Lβ-环糊精溶液(25∶75),检测波长为205 nm,流速为1.0 mL/min。结果积雪草苷和羟基积雪草苷质量浓度线性范围分别为16.86~101.18,17.89~107.33μg/mL,平均回收率分别为97.36%和97.99%,RSD分别为1.65%和1.43%。结论高效液相色谱法灵敏、准确、重现性好,比薄层扫描法能更好地控制三金胶囊的质量。
OBJECTIVE To establish a HPLC method for the determination of asiaticoside in Sanjin Capsule instead of the original TLC method to better control the quality of the preparation. Methods The column was Spherisorb ODS C18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile-2 mmol / L β-cyclodextrin (25:75). The detection wavelength was 205 nm and the flow rate was 1.0 mL / min. Results The linear ranges of asiaticoside and asiaticostigmine were 16.86 ~ 101.18 and 17.89 ~ 107.33μg / mL, respectively. The average recoveries were 97.36% and 97.99% respectively, with RSDs of 1.65% and 1.43%, respectively. Conclusion High performance liquid chromatography is sensitive, accurate and reproducible. Compared with TLC, the quality of Sanjin capsule can be controlled better.
