目的:探讨益气活血开窍复方脑脉康对缺血再灌注大鼠脑保护作用的机理。方法:以线栓法制成大鼠脑缺血再灌注模型,随机分为正常组、模型组、预防组、治疗组,后两组给予脑脉康混悬液灌胃,采用全自动氨基酸分析仪、干湿重法、原子吸收分光光度计测定脑组织中兴奋性氨基酸(EAA)含量、含水量及钙离子(Ca~(2+))含量,HE 染色观察脑组织形态学改变。结果:(1)中药组谷氨酸(Glu)含量下降(P<0.01),谷氨酸/γ-氨基丁酸(Glu/GABA)比值趋向于正常,脑组织含水量及Ca~(2+)含量下降(P<0.05)。(2)中药可减轻由于缺血再灌注所导致的脑细胞形态学的改变。结论:益气活血开窍复方脑脉康颗粒通过抑制 EAA 释放、Ca~(2+)内流而发挥脑保护作用。
Objective: To investigate the protective mechanism of Yiqi Huoxue Kaiqiao compound fangmai mai on brain protection from ischemia-reperfusion in rats. METHODS: The rat model of cerebral ischemia-reperfusion was made by the method of thread embolism. The rats were randomly divided into normal group, model group, prevention group and treatment group. The latter two groups were given Naomaixang suspension by gavage, and the automatic amino acid analyzer was used. The content of excitatory amino acid (EAA), water content and calcium ion (Ca 2+ ) content in brain tissue were measured by dry weight and wet weight method and atomic absorption spectrophotometer. HE staining was used to observe the morphological changes of brain tissue. Results: (1) The glutamate (Glu) content in the Chinese herbs group decreased (P<0.01), the ratio of glutamic acid/gamma-aminobutyric acid (Glu/GABA) tended to be normal, the brain tissue water content and Ca~(2+) ) The content decreased (P<0.05). (2) Chinese herbs can reduce the changes of brain cell morphology caused by ischemia-reperfusion. Conclusion: Yiqihuoxue Kaiqiao Compound Fangnaoxion Granule exerts brain protection by inhibiting EAA release and Ca~(2+) influx.
