目的:研究藿香正气液对湿阻证大鼠抗氧化作用及对胃黏膜表皮生长因子受体(EGFR)表达的影响。方法:将24只SD大鼠随机分为正常组、模型组、藿香正气液组,每组8只。正常组常规喂养,其余各组采用改进的环境加疲劳法制造湿阻证模型,连续造模6 d。造模成功后,正常组和模型组予生理盐水ig(20 mL.kg-1),治疗组予藿香正气液ig(20 mL.kg-1),1次/d,连续8 d。采用比色法检测血清中褪黑素(MT)、谷胱甘肽过氧化物酶(GSH-Px)及丙二醛(MDA)的含量变化,并用免疫组化方法观察胃黏膜EGFR表达的情况。结果:与正常组相比,模型组大鼠脾虚症状明显,胃黏膜有明显充血水肿,并有出血点及血块,血清MT及GSH-Px含量降低(P<0.05),MDA含量增高(P<0.05);经藿香正气液治疗后,与模型组比较,血清MT及GSH-Px含量增高(P<0.05),而MDA(P<0.05)含量降低;免疫组化结果显示模型组大鼠胃黏膜EGFR表达减少(P<0.05),藿香正气液组则明显增加(P<0.05)。结论:藿香正气液可明显改善湿阻证大鼠脾虚症状并对胃黏膜损伤有保护作用,其机制可能与增强机体抗氧化应激能力及提高胃黏膜EGFR的表达量有关。
Objective: To study the effects of Huoxiangzhengqiye on the anti-oxidation and epidermal growth factor receptor (EGFR) expression of gastric mucosa in rats of damp-resistance syndrome. Methods: Twenty-four SD rats were randomly divided into normal group, model group and Huoxiang Zhengqiye group, with 8 rats in each group. The rats in the normal group were fed routinely. The rest groups used the improved environment plus fatigue method to make the model of damp-proofing and established 6 consecutive days. After successful modeling, normal and model groups were treated with ig (20 mL.kg-1) normal saline ig (20 mL.kg-1) once a day for 8 days. The levels of serum melatonin (MT), glutathione peroxidase (GSH-Px) and malondialdehyde (MDA) were measured by colorimetric method. The expression of EGFR in gastric mucosa was observed by immunohistochemistry . Results: Compared with normal group, the symptoms of splenic asthenia in model group were obvious, the congestive and edema of gastric mucosa were obvious, bleeding points and blood clots were found, the contents of MT and GSH-Px in serum were decreased (P <0.05) 0.05). Compared with model group, the contents of MT and GSH-Px in serum increased (P <0.05) and the content of MDA (P <0.05) decreased in Huoxiangzhengqi liquid group. The results of immunohistochemistry showed that the content of MDA Mucosal EGFR expression decreased (P <0.05), Huoxiangzhengqi liquid group was significantly increased (P <0.05). Conclusion: Huoxiangzhengqi Liquid can obviously improve the symptoms of splenic asthenia in rats with dampness syndrome and protect gastric mucosal injury. The mechanism may be related to enhancing the antioxidant capacity and increasing the expression of EGFR in gastric mucosa.
