目的 应用微矩阵表达谱基因芯片研究食管癌的基因变化。方法 按一步法抽提三例食管癌及其对照癌旁组织的总RNA ,分别逆转录成荧光分子掺入的探针 ,混合后杂交基因芯片(1 4 1 1 4种基因 )。经严格洗片后用扫描仪扫描芯片荧光信号图像 ,计算机分析后比较两种组织中差异表达的基因。结果 在所检测的 3例临床标本中 ,共有的差异表达基因 31条 ,上调基因 1 0条 ,下调基因 2 1条 ,有多类基因 ,有与信号转导有关、与细胞周期调控有关、与癌基因同源的基因等等。结论 食管癌和对照癌旁组织间存在差异表达的基因
Objective To study the genetic changes of esophageal cancer using microarray gene microarray. Methods Total RNA was extracted from three cases of esophageal carcinoma and its adjacent tissues by one-step method. The total RNA was reverse transcribed into fluorescent probes and hybridized cDNA microarray (11414 genes) respectively. After the rigorous wash, the scanner scanned the fluorescence signal of the chip, and then compared the differentially expressed genes in the two tissues after computer analysis. Results There were 31 differentially expressed genes, 10 up-regulated genes and 21 down-regulated genes in 3 clinical samples tested. There were many kinds of genes involved in signal transduction and cell cycle regulation. Oncogene homologous genes and so on. Conclusion There are differentially expressed genes in esophageal cancer and adjacent tissues
