[目的]通过实验探索并验证行之有效的体外骨髓间充质干细胞分离、纯化、培养的方法。[方法]采用密度梯度离心法和贴壁培养法相结合的方法对骨髓间充质干细胞进行体外培养。[结果]原代细胞10d左右达85%以上融合,传代后细胞约1周左右达85%以上融合;传至5代细胞形态呈较一致的长梭形。[结论]运用贴壁培养法和密度梯度离心法相结合的方法对骨髓间充质干细胞进行体外培养,可获得足够数量纯度较高的骨髓间充质干细胞。
[Objective] To explore and validate the method of isolation, purification and culture of in vitro mesenchymal stem cells in vitro. [Method] The bone marrow mesenchymal stem cells were cultured in vitro by density gradient centrifugation method and adherent culture method. [Result] The primary cells reached more than 85% fusion around 10 days, and the cells reached more than 85% after about 1 week of passage. The cells of the 5th generation reached the same long fusiform shape. [Conclusion] The bone marrow mesenchymal stem cells cultured in vitro and in combination with density gradient centrifugation method can obtain a sufficient number of high purity bone marrow mesenchymal stem cells.
