目的观察galectin-1对LoVo细胞生长的影响。方法构建galectin-1真核表达载体转染LoVo细胞,观察LoVo细胞生长情况。用免疫细胞化学方法检测转染后细胞galectin-1蛋白表达水平,免疫印迹检测转染后细胞Bcl-2表达的变化。结果成功构建了galectin-1真核表达载体pEGFP-C1/GAL1(p-GAL1),并建立了稳定的空载体(LoVo)细胞和真核表达载体转染细胞(p-LoVo细胞和p-GAL1-LoVo细胞)。p-GAL1细胞可表达galectin-1,而另2组细胞则不能。galectin-1表达可降低LoVo细胞Bcl-2的表达,诱导的细胞凋亡增加,3组细胞凋亡率分别为(9.61±0.56)%,(3.56±0.53)%和(3.46±0.46)%(P<0.001)。而细胞生长曲线表明,p-GAL1-LoVo细胞增殖与p-LoVo和LoVo细胞相似。结论galectin-1可促进LoVo细胞凋亡可能与galectin-1可降低LoVo细胞Bcl-2的表达有关。
Objective To observe the effect of galectin-1 on the growth of LoVo cells. Methods The galectin-1 eukaryotic expression vector was constructed and transfected into LoVo cells to observe the growth of LoVo cells. The expression of galectin-1 protein was detected by immunocytochemistry, and the expression of Bcl-2 was detected by Western blotting. Results The galectin-1 eukaryotic expression vector pEGFP-C1 / GAL1 (p-GAL1) was successfully constructed and stable LoVo cells and eukaryotic expression vector transfected cells (p-LoVo cells and p-GAL1 -LoVo cells). p-GAL1 cells express galectin-1, while the other two groups do not. The expression of galectin-1 decreased the expression of Bcl-2 in LoVo cells, and the apoptotic rates of the three groups were (9.61 ± 0.56)%, (3.56 ± 0.53)% and (3.46 ± 0.46)%, respectively P <0.001). The cell growth curve showed that the proliferation of p-GAL1-LoVo cells was similar to that of p-LoVo and LoVo cells. Conclusion galectin-1 can promote LoVo cell apoptosis may be related to galectin-1 can reduce LoVo cells Bcl-2 expression.
