在酵母Candidautilis细胞壁上发现一种细胞壁主要结构蛋白(CWP33),其分子量为33×103,与其它细胞壁蛋白不同,该蛋白在细胞壁上对胰蛋白酶作用不敏感,而其它细胞壁蛋白组分几乎完全被胰蛋白酶降解.利用这一性质将该蛋白从细胞壁上抽提后,经DEAE-纤维素离子交换柱层析和Sepharose凝胶层析得到纯化.经研究CWP33蛋白具有葡聚糖内切酶活性,可以水解地衣糖,但不能水解对硝基苯葡萄糖苷.该蛋白可能具有重要的结构功能.
A major cell-wall structural protein (CWP33) was found on the yeast cell wall of Candidautilis, with a molecular weight of 33x103. Unlike other cell wall proteins, this protein is not sensitive to trypsin on the cell wall, whereas other cell wall protein components are almost completely Trypsin degradation. Using this property, the protein was extracted from the cell wall and purified by DEAE-cellulose ion exchange column chromatography and Sepharose gel chromatography. The CWP33 protein has endoglucanase activity, can hydrolyze lichen sugar, but can not hydrolyze p-nitrophenyl glucoside. This protein may have important structural functions.
