目的研究人乳铁蛋白对X射线照射所致小鼠急性辐射损伤的影响。方法无特定病原体级Balb/c雄性小鼠51只,随机分成3组,即对照组、辐射组和乳铁蛋白组。辐射组和乳铁蛋白组小鼠给予2.0 Gy X射线全身均匀照射,对照组不予照射。照射后2 h,乳铁蛋白组每只腹腔注射人乳铁蛋白4.0 mg;对照组和辐射组小鼠腹腔注射生理氯化钠溶液各1.0 ml。观察人乳铁蛋白对腹腔巨噬细胞吞噬指数和吞噬百分率、周围血象、血清超氧化物歧化酶(SOD)活力、丙二醛(MDA)水平和脏器指数的影响。结果与辐射组比较,乳铁蛋白组小鼠腹腔巨噬细胞吞噬指数和吞噬百分率、血清SOD活力和脾脏指数均显著升高(P<0.05,P<0.01),血清MDA水平显著降低(P<0.01);周围血白细胞计数、红细胞计数、血小板和胸腺、肝脏指数比较,差异无统计学意义(P>0.05)。结论人乳铁蛋白对小鼠急性辐射损伤具有一定的保护作用。
Objective To study the effects of human lactoferrin on acute radiation injury induced by X-ray irradiation in mice. Methods 51 pathogen-free Balb / c male mice were randomly divided into 3 groups: control group, radiation group and lactoferrin group. Radiation group and lactoferrin group mice given 2.0 Gy X-ray whole body uniform irradiation, the control group without irradiation. Two hours after irradiation, lactoferrin was intraperitoneally injected with 4.0 mg of human lactoferrin. The control and radiation groups were intraperitoneally injected with 1.0 ml of physiological sodium chloride solution. The effects of human lactoferrin on phagocytic index and phagocytic percentage, perihepatic blood, serum superoxide dismutase (SOD) activity, malondialdehyde (MDA) level and organ index of peritoneal macrophages were observed. Results Compared with the radiation group, the phagocytic index and phagocytic percentage of peritoneal macrophages, serum SOD activity and spleen index of lactoferrin group were significantly increased (P <0.05, P <0.01), serum MDA level was significantly lower (P < 0.01). There was no significant difference in peripheral white blood cell count, erythrocyte count, platelet and thymus, and liver index (P> 0.05). Conclusion Human lactoferrin has protective effect on acute radiation injury in mice.
