目的:建立酮康唑和益康唑对映体的毛细管电泳手性拆分法。方法:10mmol.L-1HS-β-环糊精为手性添加剂,以25mmol.L-1的磷酸缓冲液,用三乙醇胺调节pH至3.5为背景电解质,运行电压为-20kV,毛细管温度为20℃,检测波长214nm,对酮康唑和益康唑对映体进行手性分离研究,并考察多种因素对分离的影响。结果:在该毛细管电泳色谱条件下,酮康唑和益康唑对映体均能得到基线分离。结论:本方法简单、快捷、经济,可适用于酮康唑和益康唑对映体手性分离。
OBJECTIVE: To establish a chiral separation method of enantiomers of ketoconazole and econazole by capillary electrophoresis. Methods: 10 mmol.L-1HS-β-cyclodextrin was used as chiral additive. The pH was adjusted to 3.5 with triethanolamine as the background electrolyte in 25 mmol·L-1 phosphate buffer. The operating voltage was -20 kV and the capillary temperature was 20 ℃, the detection wavelength was 214nm, the enantiomers of ketoconazole and econazole were studied by chiral separation, and the effects of various factors on the separation were investigated. Results: Under the conditions of capillary electrophoresis, the enantiomers of ketoconazole and econazole can be separated by baseline. Conclusion: The method is simple, rapid and economical and can be applied to the enantiomeric separation of ketoconazole and econazole.
