目的:模拟临床中老年女性绝经后合并骨关节炎的病理生理特点,建立膝骨关节炎合并骨质疏松症大鼠动物模型。方法:采用经典去卵巢法构建骨质疏松症模型后的大鼠加疲劳性跑台训练。18只6月龄雌性SD大鼠,采用抽签法随机均分为空白对照组(正常组)、单纯去卵巢组(骨质疏松组)和膝骨关节炎合并骨质疏松组(模型组),每组6只。单纯去卵巢组和模型组在无菌条件下摘除双侧卵巢后,单纯去卵巢组常规喂养,模型组术后恢复1周后行跑台疲劳性训练4周,空白组不作任何处理。5周后取右侧股骨、左侧膝关节、血清及尿液行组织学、力学及免疫学分析。结果:与空白组比较,单纯去卵巢组与模型组血清雌二醇浓度显著降低(P<0.01),最大载荷、断裂载荷降低(P<0.05);与单纯去卵巢组比较,模型组大鼠尿液CTX-Ⅱ浓度明显升高(P<0.01),模型组大鼠关节软骨呈现轻度软骨损伤。结论:采用经典去卵巢法能构建骨质疏松症模型,并在此基础上使用动物跑台进行疲劳性运动可以造成膝关节劳损,从而成功建立符合女性中老年膝骨关节炎合并骨质疏松症的新模型。
Objective: To simulate the pathophysiological characteristics of postmenopausal and postmenopausal women with osteoarthritis in middle-aged and elderly women, and to establish an animal model of osteoarthritis of the knee combined with osteoporosis. Methods: A classic ovariectomized rat model of osteoporosis was used to train fatigued treadmill. Eighteen 6-month-old female Sprague-Dawley rats were randomly divided into blank control group (normal group), simple ovariectomized group (osteoporosis group) and knee osteoarthritis combined osteoporosis group (model group) 6 per group After ovariectomized and ovariectomized, ovariectomized ovariectomized rats were reared in the ovariectomized and ovariectomized groups, respectively. After a week of recovery in the model group, treadmill fatigue training was performed for 4 weeks without any treatment in the blank group. After 5 weeks, the right femur, left knee, serum and urine were collected for histological, mechanical and immunological analysis. Results: Compared with the blank group, the serum estradiol concentration in the ovariectomized and untreated groups was significantly decreased (P <0.01) and the maximum load and fracture load decreased (P <0.05). Compared with the simple ovariectomized group, the model rats Urine CTX-Ⅱ concentration was significantly increased (P <0.01), the model group rats showed mild articular cartilage injury. CONCLUSIONS: Classical ovariectomial method can be used to construct osteoporosis model. On the basis of this, the use of animal treadmill for fatigue exercise can cause strain to the knee joint, thus successfully establishing osteoporosis in women with middle-aged and elderly knee osteoarthritis New model
