为了深入研究GhCOMT1基因的功能,确定GhCOMT1基因对棉纤维细胞壁增厚的影响。本实验通过构建棉花木质素合成途径中的咖啡酸-O-甲基转移酶基因GhCOMT1的瞬时表达载体,用将pRTL2-GUS/NIa瞬时表达空载体连接GhCOMT1基因的目的片段,获得pGUS-GhCOMT1融合表达载体,并将其转化到棉花胚珠表皮细胞中进行表达。通过检测可以发现pGUS-GhCOMT1融合表达载体在洋葱表皮细胞及棉花胚珠表皮细胞中高效表达,表明棉花GhCOMT1基因对棉纤维的的伸长有显著的影响,同时为研究棉花纤维发育调控的分子机理提供理论基础。
In order to further study the function of GhCOMT1 gene, we determined the effect of GhCOMT1 gene on cotton fiber cell wall thickening.
