目的:观察丹参注射液含药血清对离体培养兔脊柱运动节段椎间盘活性及功能的影响。方法:将14只新西兰白兔随机分为空白组与含药血清组,各7只,处死后在无菌条件下取出脊柱运动节段各25个,其中空白组含有10%无药血清,含药血清组含有10%丹参注射液含药血清,两组于培养前及培养后第3、7、14、21天,各取5个椎间盘分别进行HE染色、Ⅱ型胶原免疫组化、蛋白多糖含量测定。结果:培养14d空白组椎间盘组织形态出现部分破坏,含药血清组组织形态维持良好,21d两组组织形态出现破坏;培养14d两组Ⅱ型胶原免疫组化染色强度无显著差异,21d时均出现显著下降(P<0.05),且空白组较含药血清组下降明显(P<0.05);培养7d两组蛋白多糖PAS/AB染色强度无降低,14d两组强度均有所减弱,21d进一步减弱,且空白组改变更为明显。结论:空白组14d以后椎间盘逐渐出现退变,21d时退变明显;含药血清组培养14d仍可维持椎间盘的活性与功能,21d时也出现退变,但退变程度不如空白组显著,提示含有丹参注射液的药物血清可以一定程度上减缓椎间盘退变。
Objective: To observe the effect of Salvia miltiorrhiza injection serum on the activity and function of intervertebral disc in isolated rabbit spinal motion segment. There was no significant difference in the intensity of immunohistochemical staining of type Ⅱ collagen between the two groups on day 14 (P <0.05). The staining intensity of PAS / AB of two groups of proteoglycan did not decrease on the 7th day, and the intensity of PAS / AB decreased on the 14th day and further weakened on the 21st day , And the blank group changed more obviously. CONCLUSION: The intervertebral discs gradually degenerated after 14 days in the blank group, and degenerated obviously on the 21st day. The serum containing serum could maintain the activity and function of the intervertebral discs 14 days after the culture, and degenerated on the 21st day, but the degeneration degree was not as significant as that of the blank group Serum containing Salvia injection can reduce disc degeneration to a certain extent.
