本文用薄层扫描法对不同采集时间(5~10月份)芫花叶中羟基芫花素进行了定量分析。为合理采集芫花叶提供了实验依据。羟基芫花素:从芫花叶中提取、分得,经化学、光谱、纯度鉴定,证明为纯品。仪器及条件:CS-930双波长薄层扫描仪(日本岛津公司),用外标法,线性参数SX=3,λ_(?)=353nm,狭缝1.2×1.2mm~2,⊿y=0.1,灵敏度:中。薄层板:硅胶G(粒度10~40μ),加0.8%CMC-Na液,调匀后铺板,105℃活化1hr。1.标准溶液的制备:精密称取羟基芫花素标准品1.0499mg,于1ml容量瓶中。加甲醇适量后滴加1滴二甲亚砜试剂水浴加热使全溶。放冷,加甲醇至刻度,摇匀,即得浓度为1.0499mg/ml的标准溶液,备用。2.样品液的制备:取5~10月份干燥芫花叶各1份,精密称取1g,置直滴式脂肪油提取器中用甲醇水浴回流提取8hr,提取液浓缩后移入50ml容量瓶中,加甲醇至刻度,即得样品溶液。3.薄层层离:将标准液和6份样品溶液(各点
In this paper, the quantitative analysis of hydroxygermanin in alfalfa leaves at different collection times (May-October) was performed using thin-layer scanning. It provides experimental basis for the reasonable collection of alfalfa leaves. Hydroxyamidin: It is extracted and separated from alfalfa leaves and certified by chemical, spectral and purity. It is proved to be pure. Equipment and conditions: CS-930 dual-wavelength thin-layer scanner (Shimadzu Corporation, Japan), external standard method, linear parameter SX=3, λ_(?)=353nm, slit 1.2×1.2mm~2, ⊿y= 0.1, sensitivity: medium. Thin plate: Silica gel G (particle size 10-40μ), 0.8% CMC-Na solution was added, and the mixture was plated and activated at 105°C for 1hr. 1. Preparation of standard solution: Accurately weighed 1.0499 mg of hydroxygermanin standard in a 1 ml volumetric flask. Add proper amount of methanol and add 1 drop of dimethyl sulfoxide reagent to water bath to make the solution completely. Let cool, add methanol to the mark, shake, to obtain a standard solution with a concentration of 1.0499 mg/ml. 2. Preparation of sample solution: Take 1 to 5 parts of each dry leaf from 1st to 5th of October, accurately weigh 1g, place it in a direct drop fat oil extractor and extract it in a methanol water bath for 8hr. The extract is concentrated and transferred to a 50ml volumetric flask. Methanol was added to the scale to obtain a sample solution. 3.
