[目的] 研究加味复元活血汤对实验大鼠肝纤维化预防与治疗作用及其机制。[方法] 应用四氯化碳诱导建立大鼠实验性肝纤维化模型,连续11周。造模同时中药组用加味复元活血汤灌胃治疗,西药对照组以秋水仙碱灌胃治疗,空白、模型组则给予等体积0 85%氯化钠液灌胃。于 11 周末检测大鼠肝功能:血清总蛋白(TP)、白蛋白(Alb) 、丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST);血清及肝组织匀浆肝纤维化指标:透明质酸(HA)、Ⅳ型胶原(C Ⅳ)、层黏连蛋白(LN)、Ⅲ型前胶原(PCⅢ);并观察肝组织病理形态学改变;免疫组化方法检测肝组织C Ⅳ、LN、转化生长因子β1(TGF β1)阳性表达。[结果] 加味复元活血汤组与模型组及西药组比较:肝功能TP、Alb明显升高,ALT、AST明显下降,血清HA、C Ⅳ明显下降,肝组织匀浆 HA、LN、PCⅢ明显下降,差异均有统计学意义(P<0 01 或<0 05) ;肝组织病理改变明显减轻;肝组织内 C Ⅳ、LN 、TGF β1 阳性面积显著降低。[结论] 加味复元活血汤能有效地改善肝脏微循环,具有防治实验大鼠肝纤维化,保护肝细胞,恢复肝功能的作用。
[Objective] To study the preventive and therapeutic effects and mechanism of Modified Fuyuan Huoxue Decoction on experimental liver fibrosis in rats. [Methods] Rat hepatic fibrosis model induced by carbon tetrachloride was established for 11 consecutive weeks. At the same time, the Chinese medicine group was treated with Jiawei Fuyuan Huoxue Decoction by intragastric administration. The western medicine control group was treated with colchicine by gavage. The blank and model group were given equal volume of 0 85% sodium chloride solution by gavage. At the end of the 11th day, liver function was measured: serum total protein (TP), albumin (Alb), alanine aminotransferase (ALT), aspartate aminotransferase (AST); serum and liver homogenate liver fibrosis Indices: Hyaluronic acid (HA), type IV collagen (C IV), laminin (LN), type III procollagen (PCIII); observation of pathological changes in liver tissue; immunohistochemical method for detection of liver tissue C IV, LN, transforming growth factor β1 (TGFβ1) positive expression. [Results]Comparison of Modified Fuyuan Huoxue Decoction group with model group and western medicine group: TP and Alb in liver function increased significantly, ALT and AST decreased significantly, serum HA and C IV decreased significantly, and liver homogenates HA, LN and PCIII decreased significantly. The difference was statistically significant (P<0 01 or <0 05); pathological changes in liver tissue were significantly reduced; the positive areas of C IV, LN, and TGF β 1 in liver tissue were significantly reduced. [Conclusion] Modified Fuyuan Huoxue decoction can effectively improve liver microcirculation, and it can prevent and treat hepatic fibrosis in experimental rats, protect hepatocytes and restore liver function.
