根据迄今报导的大量有关文献,对一日内何时用抗癌药疗效最佳、毒性最低进行了评述。一些证据提示,倘若一日内以宿主24小时节奏与肿瘤组织敏感性协调,在肿瘤对特殊药物最大敏感性相关的时机给药则能造成最大平均瘤细胞杀灭,从而获最理想疗效。临床前和临床实验表明,一日内选好时机给化疗药对改善常见癌瘤治疗学指标具有实际重要意义。 动物实验证明,每日休息(即睡眠)时给鼠固定剂量阿糖胞苷(Ara-C)毒性最小,鼠死亡率15%;相比之下活动时用药死亡率则为75%。如此表明Ara-C毒性最低的时间与抗肿瘤活性最高的时间一致。
According to a large number of related literatures reported so far, the best anticancer drugs used in the first day and the lowest toxicity were reviewed. Some evidence suggests that if the 24 hour rhythm of the host is coordinated with the sensitivity of the tumor tissue within a day, the maximum mean tumor cell killing can be achieved at the time when the tumor is associated with the maximal sensitivity of the particular drug, thereby obtaining the best therapeutic effect. Preclinical and clinical trials have shown that it is of great practical significance to choose the right timing for chemotherapeutic drugs to improve the therapeutic indicators of common cancers. Animal experiments have shown that the daily dose of Ara-C (Ara-C) is the least toxic and the rat mortality rate is 15% at daily rest (ie, sleep); compared with 75%, the activity mortality is comparable. This shows that the lowest time for Ara-C toxicity is consistent with the highest time for anti-tumor activity.
